At Commercial Road, the route turns east and passes the Alfred Hospital before continuing through Prahran. It serves Prahran Market and Chapel Street, then follows Malvern Road through Toorak and Malvern.
At Burke Road, Line 72 turns north and serves Gardiner Station before continuing through Glen Iris and Camberwell. It passes Camberwell Junction and Camberwell Station before terminating at Cotham Rd/Burke Rd.
