GLNELG connects the Glenelg seafront with central Adelaide and the Royal Adelaide Hospital. It runs seven days a week through the southern suburbs, serving destinations including Morphettville, Marion Road, South Road and Victoria Square. Most journeys terminate at Royal Adelaide Hospital, while selected services continue to Entertainment Centre.
The route has its origins in the Adelaide–Glenelg railway, which opened in 1873 and was converted to an electric tramway in 1929. Later extensions took trams through the city centre and west to the hospital and Entertainment Centre. Travel is free along Jetty Road in Glenelg and from South Terrace through the city; a fare is required between Brighton Road and South Terrace.

From Moseley Square, trams follow Jetty Road towards Brighton Road. The city-bound platform is named Jetty Rd – North side; trams heading towards Moseley Square use the platform named Jetty Rd – South side. The route then passes through Glenelg East and Glengowrie, calls at the new Morphettville stop beside the racecourse, and continues through Plympton Park towards Marion Road.
The line serves South Plympton, Glandore and South Road before continuing through Black Forest, Forestville and Wayville. After Greenhill Road, it enters the city at South Terrace and runs north via City South, Victoria Square, Pirie Street and Rundle Mall. It then turns west along North Terrace, stopping at Adelaide Railway Station and City West before reaching Royal Adelaide Hospital, the usual terminus.
Selected journeys continue beyond the hospital along Port Road. They stop at Thebarton and Bonython Park before terminating at Entertainment Centre, where the stop also serves the venue's Park 'n' Ride facility.
