Line T2, known as Vespucci, connects Florence's Amerigo Vespucci Airport with Piazza San Marco. It serves Novoli, Santa Maria Novella railway station and the Fortezza da Basso.
The airport-to-Santa Maria Novella section opened on 11 February 2019. The extension to San Marco opened on 25 January 2025. The two directions use different streets near San Marco: trams towards San Marco call at Cavour, while trams towards the airport call at La Marmora - Orto Botanico. Selected early and late journeys start or terminate at Unità instead of San Marco - Università.

From Peretola Aeroporto, trams pass through Guidoni and follow Via di Novoli, serving the residential district and the university campus at San Donato. The route crosses the Mugnone on the tram bridge and continues through Buonsignori and Ponte all'Asse to Viale Redi.
After Redi, the tracks pass through Palazzo Mazzoni and emerge at Belfiore. Trams continue through Rosselli to Santa Maria Novella station, serving Alamanni and Valfonda, then run past the Fortezza da Basso.
The route follows Viale Lavagnini to Piazza della Libertà. Towards San Marco, trams continue along Via Cavour to the terminus in Piazza San Marco. On the return journey, they travel via Via La Pira and Via La Marmora before rejoining the outward route.

Buonsignori - Liceo Da Vinci is a tram stop on Via Stefano Buonsignori, next to Ponte Mahatma Gandhi. The stop is named after Stefano Buonsignori, the Medici court cosmographer and is beside the Leonardo da Vinci secondary school.
Ponte all’Asse is a tram stop on Via Luigi Gordigiani, next to the Ponte all’Asse aftger which it is named.
Redi is a tram stop beside Vle Francesco Redi, near the junction with Via S. Iacopino. Both the stop and the road are named after Francesco Redi, the 17th-century physician and naturalist.
Belfiore is a tram stop on Viale Belfiore, next to Palazzo Mazzoni. The stop is named after the area of Mantua where Italian patriots known as the Martyrs of Belfiore were executed in the 1850s.
