Line 6 of the Melbourne tram system runs between Moreland Station to the north of the city and Malvern Road & High Street, which is located in Glen Iris in the east of the city. The entire journey takes about 1 hour 20 minutes and there are trams every 5 or 10 minutes during the week and every 12 minutes at weekends.
After leaving the terminus stop Moreland Station, the line runs east along Moreland Road, stopping at Sydney Road & Moreland Road, De Carle Street & Moreland Road and Barrow Street & Moreland Road. After leaving Barrow Street & Moreland Road, the line continues along Moreland Road until it reaches until it reaches Moreland Road & Nicholson Street, where it turns right onto Holmes Street and runs south, passing Mitchell Street & Holmes Street and Albion Street & Holmes Street.

After leaving Flinders Street Station, the line contines south, crossing the Yarra River on Princess Bridge and joining St Kilda Road to stop at Arts Precinct. After leaving Arts Precinct, the line continues south along St Kilda Road, stopping at Grant Street - Police Memorial, Shrine of Remembrance, Park Street & St Kilda Road, Toorak Road & St Kilda Road, Arthur Street & St Kilda Road, Leopold Street & St Kilda Road, Commercial Road & St Kilda Road, Moubray Street & St Kilda Road and High Street & St Kilda Road.
From High Street & St Kilda Road, the line turns left onto High Street and runs east, calling at Punt Road & High Street, Perth Street & High Street and Prahran Station.
