After leaving the terminus stop Balwyn Road & Doncaster Road, the line runs west along Doncaster Road, calling at Dight Avenue & Doncaster Road, Buchanan Avenue & Doncaster Road, Osburn Avenue & Doncaster Road, North Balwyn Shopping Centre, Wattle Avenue & Doncaster Road and Burke Road & Doncaster Road. After leaving Burke Road & Doncaster Road, the line heads southwest along High Street, calling at Kew High School & High Street, Woodlands Avenue & High Street, Station Street & High Street and Harp Village & High Street.
From Harp Village & High Street, the line continues southwest along High Street, calling at Harp Road & High Street, Victoria Park & High Street, Kew Cemetery & High Street, Park Hill Road & High Street, Charles Street & High Street, Union Street & High Street, Cotham Road & High Street and Kew Junction & High Street.
After leaving Kew Junction & High Street, the High Street runs in a more southerly direction and the tram line follows it, calling at Stevenson Street & High Street and Barkers Road & High Street. Just after Barkers Road & High Street, High Street becomes Church Street and the line continues to follow it, calling at Brook Street & Church Street and Hill Street & Church Street. From Hill Street & Church Street, the line continues down High Street until it reaches the junction with Burwood Road and Bridge Road, where it turns right onto Bridge Road, stopping at Hawthorn Bridge & Bridge Road, before crossing the Yarra River.
After crossing the Yarra River, the line runs west along Bridge Road, calling at Westbank Tce & Bridge Road, Burnley Street & Bridge Road, Coppin Street & Bridge Road, Richmond Town Hall & Bridge Road, Church Street & Bridge Road, Waltham Street & Bridge Road, Epworth Hospital & Bridge Road and Punt Road & Bridge Road. After leaving Punt Road & Bridge Road, the line continues west along Wellington Parade, calling at Simpson Street & Wellington Parade, Jolimont Station/MCG & Wellington Parade, Jolimont Road & Wellington Parade and Lansdowne Street & Wellington Parade, until it reaches Spring Street, which it turns right on to and runs in a more northerly direction, until it reaches Collins Street, which it turns left on to.
Running along Collins Street, the line stops at Spring Street & Collins Street, 101 Collins Street, Town Hall & Collins Street, Elizabeth Street & Collins Street and William Street & Collins Street.
